BmNPV Affecting the Activity and Gene Expression of Phenoloxidase in Bombyx mori

Abstract: In order to clarify the effect of BmNPV infection on the activity and gene expression of phenoloxidase in hemolymph of Bombyx mori, the real-time fluorescent quantitative PCR was applied to analyze the expression of prophenoloxidase gene PPO1 and PPO2 in hemolymph of the 5th instar larvae of silkworm, meanwhile the activity of phenoloxidase was determined after feeding Bombyx mori nucleopolyhedrovirus. The results revealed that the expression of PPO1 and PPO2 presented an increasing trend after oral infected by BmNPV after 6 h and 9 h, but the relative expression quantity of PPO1 had significant difference with that of PPO2 at 6 h and 9 h. In addition, the phenoloxidase activities of the treatment group were significantly higher than that of the control group at 6 h and 9 h, while phenoloxidase activity of the treatment group was significantly lower at 24 h. As a kind of humoral immunity factor, phenoloxidase may be involved in the immune defense response of silkworm by BmNPV infection.
