Anti-browning Agents Affecting Callus Subculture of Three Legume Forage Species

Abstract: To solve the browning in callus subculture of three legume forage species, callus ofMedicago sativa ‘Xinmu No.4’,Onobrychis viciaeflia andTrifolium repnes‘Haifa’were taken as test materials, the effect of different measures on preventing browning for the three legume forage species in callus subculture were discussed. The results showed that in callus subculture of M. sativa‘Xinmu No.4’and O. viciaeflia , theinhibition effect of 0.02 g/L AgNO3 was better and the browning rate was 13% and 15%, respectively, the browning degree of callus was PVP> citric acid> AgNO3; the inhibition effect of 0.015 g/L AgNO3 was better and the browning rate was 32% during callus subculture ofT. repnes‘Haifa’, the browning degree of callus was citric acid> PVP> AgNO3. A certain concentration of AgNO3 could inhibit browning as well as promote the growth of callus to some extent.
