The Proficiency Testing Results of Pathogen Detection Ability for Koi Herpesvirus Disease (KHVD)

Abstract: To improve the overall level of pathogen detection of Koi herpesvirus disease (KHVD) in China, and strengthen the detection ability of laboratories for Koi herpesvirus (KHV), the proficiency testing programs were carried out for 3 times from 2014 to 2016. Blind samples were used in the proficiency testing programs, which were prepared by mixing carp tissue homogenate with recombinant plasmids containing KHVTK gene fragment and Sph gene fragment. The homogeneity and stability of the blind samples were examined. Polymerase Chain Reaction (PCR) assay, which was recommended by the aquaculture industry standard, Detection Methods of Herpesvirus (CyHv)-Part I: Koi herpevirus (SC/T 7212.1-2011), was used to detect KHV, and the test results of all participating laboratories were analyzed. The satisfaction rate of the first round test results was 58% in 2014, after the second round test, the overall satisfaction rate was 96%. In 2015, the satisfaction rate of the first round test results was 77%, the overall satisfaction rate was 89% after the second round tests. The satisfaction rate of the first round tests results was 82.69% in 2016, and the overall satisfaction rate was 92.73% after the second round tests. Through the proficiency testing programs of 2014-2016, most attended laboratories possessed the ability of PCR detection of Koi herpesvirus and the capacity of quality management, and could undertake the detection and monitoring of Koi herpesvirus.
