Chlorogenic Acid and Rutin from Inferior and Waste Tobacco: Extraction Process

Abstract: The paper aims to find an appropriate extraction process of chlorogenic acid and rutin from inferior and waste tobacco to improve its economic added value. The authors adopted the single factor experiment and response surface design, detected extraction yield by the high performance liquid chromatography (HPLC), optimized the parameters of the extraction process by ethanol concentration, liquid-solid ratio, extraction time and extraction temperature. The results showed that: the optimized condition for extraction of chlorogenic acid and rutin from inferior and waste tobacco was ethanol concentration of 67%, liquid-solid ratio of 1:16 (g:mL), extraction temperature of 61℃ and the extraction time of 150 min. Under this modified condition, the sum of the theoretical extraction yield of chlorogenic acid and rutin was 43.59 mg/g, and the relative error was 0.36%. Analysis of the Design-Expert showed that: ethanol concentration>extraction time>liquid-solid ratio>extraction temperature. The technological condition of extracting chlorogenic acid and rutin from inferior and waste tobacco obtained by the study could provide a new way for better utilization of waste tobacco.
