Hydrogen Sulfide: Effect on Growth, Accumulation of Flavonoid and Polyphenol in Betula platyphylla Suspension Cells

Abstract: To investigate the effects of hydrogen sulfide (H2S) on the growth and the accumulation of flavonoid and polyphenol in Betula platyphylla suspension cells, the hydrogen sulfide donor sodium hydrosulfide (NaHS) is added to the suspension culture system of Betula platyphylla for 8 days. pH meter, conductivity meter and ultraviolet spectrophotometer are used to analyze the pH value, conductivity, cell viability, malondialdehyde, catalase (CAT) and phenylalanine ammonia lyase (PAL) activity and the contents of flavonoids and polyphenols. After NaHS treatment, the pH value in Betula platyphylla suspension cells decreases, and the conductivity increases, the content of malondialdehyde, CAT and PAL enzyme activity, the content of flavonoids and polyphenols in the suspension cells of Betula platyphylla increase, while the cell viability basically declines. The NaHS treatment has a concentration and time effect on the growth of the suspension cells, and the contents of flavonoids and polyphenols. The contents of flavonoids and polyphenols are the highest with 1 mmol/L NaHS treatment for 24 h, which is 128.57% and 136.89% higher than that of the control, respectively. NaHS treatment could promote the accumulation of flavonoid and polyphenol in the suspension cells of Betula platyphylla.
