Screening and Optimization of Immobilization Methods for Apergillus oryzae HDF-7

Abstract: Apergillus oryzae immobilization enzyme production and its application are research hotspots, and screening a method qualified to the requirement of immobilization is essential. A. oryzae HDF-7 isolated from the conventional bean paste is used as a starting strain, and the effects of each single factor on the retention rate of immobilized mycotoxins are investigated. The optimal combination of each single factor is determined by orthogonal design. The results showed that the optimum immobilization method was sodium alginate method, the highest retention rate of protease was (27.77±0.80)% when the concentration of sodium alginate was 2% , the concentration of bacterial cells was 1:15, the immobilization time was 2.5 h, and CaCl2 concentration was 3%, the orthogonal protease activity retention rate was the highest of 38.67%. In this study, the best immobilization method is screened from A. oryzae HDF-7 cells and optimized to determine the optimal fermentation condition, which can be applied to solid-state fermentation.
