Optimization of Suspension Culture Condition for Producing Rosmarinic Acid from Perilla Cells

Abstract: [Objective]With perilla (Perilla frutescens) callus as raw material, the cell suspension culture conditions for the production of rosmarinic acid were studied. [Method]The effects of culture time, initial inoculum amount, hormone concentration and phenylalanine concentration on the content of rosmarinic acid were investigated by single-factor tests. The optimal culture conditions were optimized by Box-Behnken Center design of response surface method, and a quadratic polynomial regression equation of the forecasting model was established. [Result][Conclusion]The results showed that the optimal conditions for the production of rosmarinic acid by suspension culture of perilla cells were as follows: 6-BA of 2.34 mg/L, NAA of 0.96 mg/L, 2, 4-D of 0.60 mg/L, phenylalanine of 0.1 g/L. Under these conditions, the rosmarinic acid content in perilla callus was up to 23.61 mg/g.
