Preliminary Establishment of Rapid Propagation of Leucanthemella linearis by Tissue Culture

Abstract: Experiments were conducted for the standardization of micropropagation and regeneration of Leucanthemella linearis, a helophyte perennial plant. Nodal segment were used as explants and they were cultured on MS medium and 1/2MS medium supplemented with different concentrations of 6-BA and NAA both in individual and in combined form for shoot inductions and the best result of primary cultural was obtained from MS medium supplemented with 6-BA and NAA at the concentrations of 0.5 mg?L-1 and 0.5 mg?L-1 respectively. The best result of subcultural was obtained from MS medium supplemented with 6-BA+NAA at the concentrations of 0.5 mg?L-1 and 0.2 mg?L-1 respectively, resulting about 9.2 multiple shoots from a single shoot tip explant. For root induction and elongation, 1/2MS medium plus NAA at the concentrations of 0.3 mg?L-1showed an adverse effect. Callus was induced from nodal segment on MS medium supplemented with 0.2 mg?L-16-BA and 0.5 mg?L-1 NAA, and plants regenerated best from these calluses on MS medium supplemented with 1.0 mg?L-1 6-BA and 0.2 mg?L-1 NAA, the ratio of regeneration was 70.00%.
