Study on the Preparation and Purification Process of Sapogenin from Gleditsia sinensis

Abstract:

To optimize the preparation process of sapogenin from Gleditsia sinensis saponin by acid hydrolysis, the effects of acid concentration, acidolysis time, acidolysis temperature, material-to-liquid ratio and acid types on the conversion of saponin into sapogenin by acid hydrolysis were investigated. The results showed that the optimum conditions were as follows: hydrochloric acid concentration of 4 mol/L, hydrolysis time of 4 h, hydrolysis temperature of 80℃, solid-liquid ratio of 1:30 (m:v), and the content of Gleditsia sinensis sapogenin product was about 65%. The sapogenin after acid hydrolysis was purified by organic solvent extraction and recrystallization. The best extraction agent was trichloroethylene, the solid-liquid ratio was 1:20 (m:v), and extraction time did not change much within 90 minutes. After one extraction and recrystallization with 60% ethanol, the purity of total sapogenin could reach 76.31%.
