Isolation and Sequence Analysis of NBS-type Resistance gene Analogues from Hainan Common Wild Rice (Oryza rufipogon Griff)

Abstract:

Degenerate primers based on conserved motif of the nucleotide binding site(NBS) region from the cloned plant disease resistance genes were used to isolate resistance gene analogues (RGAs) from genomic DNA of Hainan common wild rice (Oryza rufipogon Griff). After T/A cloning, sequencing and analyzing by alignment, 4 RGAs with uninterrupted open reading frames (ORFs) were obtained. Sequence identity among the 4 RGAs nucleotide sequences ranged from 53.14% to 99.81%, while the 4 RGAs deduced amino acid sequences showed identity ranged from 39.08% to 99.42%. The deduced amino acid sequences of RGAs contain the conserved domains such as P-loop, Kinase-2, Kinase-3a and GLPL, and the 4 RGAs all belonged to nonTIR-NBS type resistance gene segments.
