Effect of different concentrations of caffeine and glutathione in the basic frozen semen dilution on quality of frozen dog sperm

Abstract:

Abstract: This experiment was aimed to improve the freeze-thawing effect of the dog’s sperm, and detect the effect of caffeine and glutathione in the basic semen dilution on guality of frozen dog sperm. Method: The Caffeine or glutathione was added into the routine semen dilution and then, the quality of frozen dog sperm was described by the following three aspects: spermatozoa progressive motility, sperm deformity rate and acrosome intactness rate. Results: After post-thawing, the quality of frozen dog semen was evaluated as following: (1) The sperm livability in the group added with 5mmol / L of caffeine was significantly higher than that of the other groups (P <0.01), and the sperm abnormality rate was significantly lower than that in the other groups(P <0.01), but while it was added with 12.5mmol / L of caffeine, the appropriate data was significantly lower than that in the control one(P <0.01). (2) The sperm livability, acrosome integrity rate, and malformation rate in the group added with 5mmol / L of glutathione, were all significantly better than that in the others (P < 0.01). Conclusion: It can be concluded that the appropriate supplement of caffeine and glutathione in the basic frozen semen dilution may improve the quality of dog sperm through their normal frozen-thawing processes.
