  Effect of different concentrations of caffeine, VE on the freezing semen of small-tailed Han sheep Cui Xu1, Luo Guihe2, Li Weidong3, Li Fuhuang4, Meng Danna2, Lü Jianwei1, Mu Di1, 2010, 26 (8):  1-6.  Abstract + ( 1387 )   PDF ( 1573914KB )   ( 1174 ) Abstract: 【Objective】This study was aimed to investigate the effect of different concentrations of caffeine, vitamin E (VE) on the freezing semen of small tailed Han sheep. 【Method】The different concentrations of caffeine and VE were added to the pre-frozen sheep semen diluent in small-tailed Han sheep, then the motility rate of their post-thawed semen, the percentage of sperms with intact acrosome, and the percentage of their abnormality, were respectively estimated to detect the quality of their frozen semen. 【Result】The results were shown as following: While the concentration of VE was 0.4mg/ml, after freezing and thawing, the livability rate of the semen from small tailed Han sheep is high, whereas their abnormality rate is low, but the apical body percentage of sperm head is still high. Meanwhile, adding with 5mg/ml caffeine in their freezing diluents, can also obviously improve their sperm livability rate(0.35), but not affect their abnormal rate and the apical body rate. 【Conclusion】Adding appropriate concentrations of vitamin E to the frozen semen diluent, can slow the freezing of sperm damage and improve small-tailed Han sheep frozen sperm viability, improve the quality of frozen semen; When adding caffeine dilution of 5mg/ml solution, after frozeing, the motility rate was significantly increased, but does not affect the sperm deformity rate and the rate of acrosome intactness.

The recombinant vectors were transformed into recipient germs BL21. Samples were collected at different induction time after induction with IPTG. The specificity of the expressed proteins were identified with SDS-PAGE, Western Blotting . And the protein were about 23kDa and 28 KDa in size. The expressed protein were purified byNi-NTA His-Bind purification system and the degree of purity is about 91%.The purified protein was coated on 96-well plate at 60μg/well and used for identification of ADV-positive sera from different regions. The results are 93% identical to CIEP. But this method is safer and easier to perform. High efficient expression of ADV protein will benefit for diagnosis of ADV in practice.

  Analysis on Correlation Between Microsatellite Marker AY493302 and Eggshell Color of Jinyun Duck Yuan Qingyan1, Tao Zhengrong1, Li Guoqin1, Shen Junda1, Chen Yichun2, Chen Wenbi 2010, 26 (8):  12-16.  Abstract + ( 1256 )   PDF ( 1600939KB )   ( 1087 ) Microsatellite locus AY493302 was used to analyze the polymorphisms of 50 blue-eggshelled and 30 white-eggshelled Jinyun ducks for the correlation relationship between this marker and eggshell color of Jinyun ducks. The results showed that the genotype distributions of this marker between two eggshell color ducks were significantly different (P=2.247E-12). This denotes that microsatellite AY493302 was highly correlated with eggshell color of Jinyun duck. The frequency of genotype JJ (365bp/365bp) was also significantly different between two eggshell color ducks (P<0.0001). Furthermore, this genotype existed only in blue-eggshelled Jinyun ducks, while not in white-eggshelled ones. These suggested that genotype JJ (365bp/365bp) can be used to select the homozygous blue-eggshelled gene of Jinyun duck in the form of assistant marker.

Control groupdiets containing extruded full-fat soybean (EFFS). The whole experiment duration was 30 days, All rats were slaughtered on 30 d. pancreas ,liver ,speen ,kidney and stomach were collected quickly for weighting . duodenum central region and jejunum1/3 and ileum1/2 were collected to making histological section for light microscope to inspected , respectively. Then, the gut measured length and weight. Results showed: The pancreas weight、the Jejunum crypt depth and The jejunum Villus height：The jejunum Crypt depth of the test groups were significantly (P<0.05) difference compared to the control group. Effect of antinutritional in feeding rats with raw sobean diets is Jinlinxiaoli6＞Jilin30＞fengjiao7607＞Jilin45＞Jinong.

  Effect of different concentrations of caffeine and glutathione in the basic frozen semen dilution on quality of frozen dog sperm Lin Zili, He Yanfang, Lu Tingting, Kang Rui, Qin Xiurui, Zhang Xin, Ni Hemin 2010, 26 (8):  21-24.  Abstract + ( 1182 )   PDF ( 1608324KB )   ( 977 ) Abstract: This experiment was aimed to improve the freeze-thawing effect of the dog’s sperm, and detect the effect of caffeine and glutathione in the basic semen dilution on guality of frozen dog sperm. Method: The Caffeine or glutathione was added into the routine semen dilution and then, the quality of frozen dog sperm was described by the following three aspects: spermatozoa progressive motility, sperm deformity rate and acrosome intactness rate. Results: After post-thawing, the quality of frozen dog semen was evaluated as following: (1) The sperm livability in the group added with 5mmol / L of caffeine was significantly higher than that of the other groups (P <0.01), and the sperm abnormality rate was significantly lower than that in the other groups(P <0.01), but while it was added with 12.5mmol / L of caffeine, the appropriate data was significantly lower than that in the control one(P <0.01). (2) The sperm livability, acrosome integrity rate, and malformation rate in the group added with 5mmol / L of glutathione, were all significantly better than that in the others (P < 0.01). Conclusion: It can be concluded that the appropriate supplement of caffeine and glutathione in the basic frozen semen dilution may improve the quality of dog sperm through their normal frozen-thawing processes.

Methods The different concentrations of ethylene glycol(2%, 4%, 6%, 8%, 10%) were added in conventional frozen diluted solution of canine semen respectively, the post-thaw semen motility, the percentage of sperms with intact acrosome and the percentage of abnormality were respectively analyzed to estimate the quality of frozen semen. Results The sperm motility in the group with 6% ethylene glycol was 0.37, and its percentage of sperms with intact acrosome was 43%, much higher than those of the other groups(P＜0.05). The percentage of abnormality of 4% ethylene glycol was 15%. Conclusion 6% of ethylene glycol could replace the glycerol as a cryoprotectant, and improve the effect of freezing canine sperm.

Put the organic iodine suppository to the uterine in the 1st,3rd,5thdays drespectively, according to 0.01g/kg weight (in terms of effective iodine) for treatment, to observe the clinical symptoms, uterine shape, pathological changes of endometrium and serum SOD, MDA, GSH-Px and other biochemical parameters before and after treatment. Results In the 4thd,vulva swelling and vaginal congestion gradually subsided ,uterine secretions gradually reduced; cervical and horn of the uterine swell slightly, a small amount of clear secretions is inside the uterine; endometrial is more complete, epithelial cell structure is normal. In the4thd, serum SOD and GSH-Px in treatment group activity were significantly higher than that in pathological control group (P<0.05). While in the 6thd, serum SOD in treatment group activity was significantly higher than that in pathological control group (P<0.05),serum GSH-Px activity was very significantly higher than that in pathological control group (P <0.01),serum MDA contents was very significantly lower than that in pathological control group (P <0.01).Conclusion The organic iodine suppository may be effectively cure the rabbit endometritis.

In this study,the sequence of VP1 ,which was isolated through RT-PCR , was analyzed in DNAstar software to abtain its spatial arrange and antigenicity . The result of this research indicated that the VP1 from OS/99 might contain many antigenic sites in terms of some parameters on the antigenicity , which located in 22-29，43-46，82-85，92-93，100-111，134-139，154-156，180-184，196-203 amino acid regions . For that , the virus strain of os/99 showed the powerful ability in aspect of more broader coverage in antigenicity Together ,it was suggested that the spatial conformation of VP1 in OS/99 virus strain ,which were subcultured for 13 generations , have not been obviously changed .

(Result)The method of rumen fermentation in vitro was used to study the effect of various concentrate supplement, the rumen micro-organisms, incubation time to rumen hydrogenation. The results showed that: NH3-N was effected mainly by different materials and the interaction between Microflora and incubation time,the content of NH3-N was significantly higher in the full-fat soybean group than the soybean baking group,but had no significant difference with health soybean. The concentration of NH3-N in the fungi and Rumen fluid medium was increased fastest and it was at the maximum position all long. VFA concentration was effected mainly by the interaction between microflora and incubation time and little by different concentrate supplement. The concentration of propionate and butyrate were higher in the group of rumen liquid. Different material, incubation time and interaction between them had significantly effcet to the content of stearic acid.the remarkable influence to the concent of stearic acid was produced only in some time in rumen microflora. Concentration of linoleic acid(18:2) was impact only by different rumen microflora. Different material, incubation time and interaction in rumen microflora had significantly effect to the concentration of trans-oleic acid (18:1) and linolenic acid (18:3). (Conclusion) Fungi in the supplementary materials play an important role in fermentation, VFA was the result of interaction between different micro-organisms,adding full-fat expanded Soybean had a positive effect on the synthetic-CLA and its precursors.

The weight of healthy rabbits was 2.5±0.25kg, they were randomly divided into 4 groups, the model group, the honeysuckle compound group and the Herba Artemisiae Scoparia group were injected the endotoxin saline with intravenous, the dose was 100μg/ kg, after 24h, the Compound Honeysuckle and Herba Artemisiae Scoparia groups were injected the Chinese medicine with intravenous, the dose was 10ml/kg. The model group was injected same volume saline intravenously. The negative control group was injected same volume saline intravenously with the endotoxin and Chinese medicine injection group, after 24 hours, jugular vein bleeding to death, take heart, lung tissue, Bouin liquid fixed 24h, made paraffin sections, stained of HE, microscope and photography. The results showed that the rabbit of model group myocardial fibers were swelling, parts of fracture, interstitial congestion, around vascular edema, the cell of myocardial had vacuolar degeneration, the dispersivity edema of lung, the small vascular hyperemia in interstitial of lung, there were the phenomenon of neutrophils accumulation in the lumen, the micro thrombus formed, around small vascular congestion; the massive inflammatory infiltration, the structure of alveolar was damaged. The rabbit of Compound Honeysuckle group myocardial fibers arranged tidiness, there were a little congestion around myocardial fibers; the most parts of alveolar septa normal, the small parts of swelling, thickening. Herba Artemisiae Scoparia group heart, lung tissue structure compared with the model group has basically not changed.

Temperature seted 30℃, 35℃, 40℃, moisture were 35% and 40%.The results were showed that there were interaction between temperature and moisture. Compared with not fermented rapeseed meal, pH and acid-binding capacity of fermented rapeseed meal-sol were significantly reduced（P<0.01）,crude protein content was significantly increased（P<0.01）.Ferment temperature and moisture were 35℃ and 40%, pH was the lowest ,crude protein content was the highest, acid-binding capacity and crude fiber was lower and had significant difference compared with not fermented rapeseed meal. Optimal ferment temperature and moisture were 35℃ and 40%.

The molecular weight of DNA, isolated by modified CTAB method, was nearly 21 kb, and the OD260/OD280 was about 1.8. Using the DNA isolated by modified CTAB method as the template in ISSR-PCR, the excellent amplification result, with distinct band and fine repeatability, could be obtained.

The results showed that the fructose content, glucose content and sucrose content showed decreasing tendency upon the whole storage method. The parameters tested of No.3 new sorghum were significantly higher than No.4 new sweet sorghum. As the storage time goes on, the content of three parameters above from highest to lowest is defoliation stand, defoliation lie, leaf stand, leaf lie, pyramid type 5th, pyramid type 4th, pyramid type 3rd, pyramid type 2nd ,pyramid type 1st in the whole storage period.

Special fingerprint of local capsicum var was constructed,which provided information for the local pepper species pretection.

Fruit setting proportion among eight hybrid combinations of wild-type tea plants and cultivar-type tea plants ranged from 4.4% to 22.5%, showed that cultival-type tea plants with high Pollen viability as fathers were more efficient than wild-type tea plants with low pollen viability. Moench) varieties, 654, ICSV111/1022 and BTx623, were irradiated by 60Co-γrays with 4 dosage levels. The morphologic and physiologic variations at germination and seedling stage were investigated and the appropriate dosage levels of 60Co-γray for different sorghum genotypes were estimated.The seed germination energy and germination rate were firstly promoted when the dosage level was lower than 300Gy，and eventually inhibited when the dosage level was increased. The seedling growth and vigor index were declined gradually with increasing dosage. The activities of protective enzymes SOD, CAT and POD were elevated gradually with increasing dosage. Significant negative correlation existed among seedling growth, vigor index and dosage, while the activity of protective enzymes had significant positive relationships with dosage. The results suggested that the interference effect could be evaluated by the parameters of germination rate, seedling growth, vigor index, SOD, CAT and POD activity in M1 plants. The irradiation endurance of 3 varieties was ICSV111/1022>654>BTx623. The suitable irradiation dosage for irradiation breeding of 3 sorghum varieties was determined within the range of 300~700Gy.

The six bamboo species are Phyllostachys aurea，Phyllostachys aureosulcata f. spectabilis， Sasa argenteastriatus，Phyllostachys arcana f.luteosulcata, Phyllostachys nidularia and Phyllostachys arcana. The results are as follows: The survival rate of Phyllostachys aurea, Phyllostachys aureosulcata f. spectabilis, Sasa argenteastriatus ,Phyllostachys arcana f.luteosulcata and Phyllostachys nidularia, which was 94.84%,87.16%,100%,100%and 95.16% respectively. The SOD of Phyllostachys arcana was steady and those of other five bamboo species were negatively correlated to the temperature. The paclobutrazol solutions were applied at full flowering stage of peanut. The results indicated that when 75-100 mg/L paclobutrazol was applied at blossoming stage, the growth of the main stems and branch stem were significantly inhibited, the amount of legume per plant and percentage of full legume were increased, and the peanut yield was also increased. Most of the growth parameters studied was affected by the concentration of paclobutrazol application. When the concentration of spraying paclobutrazol was increase among 0-100 mg?L-1, it would result in the significant increase of the peanut yield. The correlation analysis showed that the linear relationship between biomass and the pod yield were PP333:y=0.0240+0.3919x（r=0.0402），CK：y=0.2587+0.2680x（r=0.0485）. The relationship between the plant height and the fresh biomass were CK:y=0.0426+0.8411x(r=0.0070), PP333y= -0.1126+9.4824x(r= -0.0045). The morphology indices augment with treatment time prolonging by all PEG-6000 grades and their difference appear after 3-4 days. CAT and SOD activity augment with treatment time prolonging and PEG-6000 concentration increasing. The resistant one augments faster than the sensitive one and descends slower than the sensitive variety. MDA and Pro content augment with treatment time prolonging and PEG-6000 concentration increasing. The effect of the sensitive variety is stronger than the resistant one. So they are not adapted to be drought resistant indices. Root activity augments with treatment time prolonging. The consistent with or coincide with or et al optimum treatment is T1. Correlation analysis showed that there were very significantly positive correlations between root activity and morphology indices, and significantly or very significantly negative correlations between root diameter, CAT activity, SOD activity and MDA content, praline content. MDA content was very significantly positively related to praline content, CAT activity was very significantly positive correlation to SOD activity.

The generation rate of the superoxide free radical (O2-?), hydrogen peroxide (H2O2) and malondialdehyde (MDA) contents, electrolytic leakage, chlorophyll contents changed slightly. No obvious injury was observed in plant growth. The results indicated that the heat tolerance of Bromeliaceae was improved by enhancing the activities of SOD, POD and CAT and the contents of Pro and HSP. The results showed that the net photosynthetic rate (Pn) and stomatal conductance (Gs) were significantly decreased after treatment of drought stress, and that intercellular CO2 concentration (Ci) was slightly decreased. These results suggested that the decreased Pn under drought stress was resulted from the stomatal and non-stomatal factors. Drought stress decreased the efficiency of excitation energy captured by PSII (F′v/F′m) and photochemical quenching (qP), thereby resulting in a decreased electron transport flow (ETR). However, the non-photochemical quenching (NPQ) and the excess energy of PSII reaction centers (Ex) were significantly increased after drought stress, which indicated that the partial excitation energy was dissipated by antenna pigments, but the excessive excitation energy could not be dissipated efficiently, thereby resulting in photoinhibition in Eremochloa ophiuroides under drought stress conditions.

Additionally, the result of this research also suggested the PCD initiates in proximal part of root under N dificiency.The results that amino acids and soluble sugar concentrations in oat root exdudation under no N condition were not significantly higher than those under abundant N supplies suggested that amino acids and soluble sugar concentrations in oat root exdudation has nothing to do with root PCD. (2) Correlation between productivity index and corn yield in common years were analyzed, The highest Determination coefficient with R2=0.6774(P<0.01) was appeared in BPI model, followed by PI model with R2=0.3285(P<0.05), the lowest was appeared in CI model (P>0.05). (3) PI model over-estimate the result for those soil with low organic content, after import organic factor in to the model, BPI got a better estimation for black soil.【CONCLUSION】In conclusion, PI model take upper 100cm horizon soil as estimated object of soil productivity, the estimated accuracy is better than CI model. If organic factor was considered, the BPI model got a better estimation than PI model. Besides the organic factor, whether the other soil physico-chemical properties is helpful to improve accuracy for BPI in North-East black soil region, it need research further in future.

No remarked differences in the activities of SOD and POD, MDA content and chlorophll content were observed between treatment N3 and N2, indicating that when the nitrogen application amount rose to a reasonable level, the flag leaf senescence can not be slowed down by continuous increase of nitrogen application. ② The process of senescence could be divided into two periods, i.e. slow-senescence phases and fast-senescence phases. In slow-senescence phases, the activities of POD and SOD, MDA content and chlorophll content in flag leaf increased slightly. At fast-senescence phases, the three physiological parameters decreased sharply, and the wheat showed obvious senescence characteristics. Thus, we separate the endogenetic microorganism from banana tree, and we find bacteria are dominating in these microorganism. In these bacterias, some secrete chitinase which can restrain the pathogen’s growth especially in culture medium made of banana lixivium. The phenomenon provide an good idea for biocontrol against Fusarium wilt.

[METHOD]Different temperatures of the developmental period, developmental rate, developmental threshold temperature, effective accumulated temperature of T. molitor were studied with effective temperature method . [RESULTS]Results indicated that the effects of temperature on T. molitor were that the temperature was higher and the developmental durations was shorter and growth rate accelerated. Pupal stage developmental threshold temperature of the highest, 11.8816 ℃, while the egg stage of the developmental threshold temperature of the lowest, 8.0021 ℃.The larval stage the maximum effective temperature for the 67.6078 ℃?d. T. molitor finished a generation with 47.02 days in 24 ℃. However, 38.01 days were needed to finished a generation in 28 ℃. [CONCLUSION] So the damage of T. molitor for rush products could be controlled effectively with low or high temperature treatment.

Genomic DNA of the sampled populations came from Fujian, Hainan, Guangdong, Yunnan and Sichuan，was extracted from thoracic tissue of individual adult ?ies by CTAB method and amplified with the polymerase chain reaction. By using microsatellite markers (SSR), the fingerprint of five geographic population was investigated. With good reproducibility and high polymorphic, seven microsatellite primers were utilized in this study to assess the genetic polymorphism of orient fruit fly primarily. The results indicated that genetic differentiation exists among the five populations.

The filtrate and crude toxins (ethyl acetate extract of culture filtrate of HGE) of HGE were tested with the method of stiletto, and measured in MIC(Minimun Inhibitory Concentration) and MBC(Minimal Bacteriocidal Concentration), compared with the checks of 20% thiodiazole-copper, 72% streptomycin sulfate and bismerthlazol. The result indicated that the biocontrol effect from the treatment of the crude toxins of HGE was much better than those of other treatments.

Through the research, we have found out the transportation ecological footprint accounted for the largest proportion of the total tourism ecological footprint which is 5739.81 hm2, the food ecological footprint is second only to it, the shopping ecological footprint and the entertainment ecological footprint is considered negligible. The ecological deficit of the whole scenic areas is 4020.13 hm2. Only if the ecological resources are exploited reasonbly, is the sustainable development of the tourism guaranteed.

A MBR consists of intermittent aeration zone and membrane separation zone with total volume of 120L. Nine tests were arranged at different HRTs(15h, 20h and 24h) and SRTs(70d, 30d and 50d) respectively. The kinetic constants were calculated using the data from experiments. A kinetic model was satisfactorily employed to predict MLVSS in reactors during experiments.

The content of Hg, Cd, Pb, As, Zn, Cu, Cr and Ni were not exceed the second class of the standard of soil environmental quality. Hg, Cd, Pb in wheat grain were not exceed the national food and healthy standard value. While As content was exceeded standard value by 0.02 mg/kg at the amount of 1800kg/hm2 level of sludge compost. Zn、 Cu content in straw was lower than grain, and other heavy metals content was 2-40 times of grain. So in a short time, proper amount of sewage sludge fertilizer will not caused pollution in soil, or bring effects in edible part, either. But years of repeated application demand timely monitoring.

The results showed that: (1) Cattle, horses require cool, humid climatic conditions; donkeys, mules, camels, goats and sheep have a strong capacity to adapt to the hot and dry climatic conditions. Camels, goats and mules have a strong capacity to adapt to the strong sunlight conditions, while sheep have the poor adaptability. Except for no significant differences between the deficient and normal plants for B content in roots and K in fruits and upper leaves, Ca and Mg in fruits and lower stems, the B and K contents in B-deficient plants were lower the normal ones for top and upper leaves, petiole, stem and fruits, even for K in roots, reversely Ca and Mg contents were higher in B-deficient plants than B-normal ones. The results show that: The probability of occurrence of low temperature and cold damage is big, but the frequency generally goes down; arid disaster and flood disaster occurs frequently in Harbin region, the strength is not strong on the whole; occurrence frequency of arid and flood disaster all have been above 65% in the past 35 years, but strong disaster is little; the average number of days of thunderstorms is 33.54d with the last 35 years of Harbin, only one year is strong disaster year with the number of days is more than 50d,the thunderstorms occur mainly at June, July and August. In the main meteorological disaster of Harbin, the probability of arid and flood disaster is the highest, followed by the low temperature and cold damage.

One medication group was injected with cyclophosphamide and astragalus injection at the same time as drug-treated control fish.The other three groups were injected with cyclophosphamide and plus Yupingfeng power simultaneously.Thirty-three eels were treated with 0.8ul/g?BW plus Yupingfeng power ,which was made up of Huangqi,Baizhu,Fangfeng,Dangshen and Gancao,as low-dosage-drug-treated fish, middle-dosage-drug-treated fish were injected with 1.6ul/g?BW plus Yupingfeng power and the high-dosage-drug-treated fish was injected with 3.2ul/g?BW plus Yupingfeng power. Counting of mortality rate and spleen index,and detecting the change of antioxidant enzyme activities at the 15th day of experiment. 【Results】The results indicated that,compared with the masculine-blank contral group, the mortality rates of each treated group were decreased,the level of spleen index was elevated,the activities of SOD,CAT and GSH-Px increased obviously,and the low-dosage-drug-treated group showed the best effect.Overall,these results as mentioned above demonstrated that the Plus Yupingfeng Power can enhance the level of spleen index,and also bring the activities of SOD,CAT and GSH-Px going up significantly. 【Conclusion】The preparation has a effective immunopotentiator against the experimental immunosuppression of Monopterus albus.

Taking 17 prefectures as samples, under the help of Matlab and MapInfo we made a correspondence analysis of rural residents ‘consumption pattern in different prefecture of Shandong province, and we got comprehensive correspondence information exist in indexes, in samples and between indexes and samples, finally we assorted 17 prefectures, autonomous regions and municipalities into 4 types, and analyzed the difference of rural residents ‘consumption pattern of every sample.

In the end, we give advice to the long-time mechanism building.
