Protective effect of Shuanghuanglian on acetaminophen injuried Canine hepatocytes

Abstract:

Canine primary cultured hepatocytes were isolated by a modified two-step collagenase digestion method. The injuried hepatocyte model was made by acetaminophen. The injuried hepatocytes were treated by Shuanghuanglian, ALT and AST activities were measured, the expression of CYP450 was determined by Western blot. The results as follows, the activities of ALT and AST in increased significantly by 16mM AP treated for 12h. The activities of ALT and AST decreased significantly. The expression of cytochrome P450 in injuried Canine hepatocytes increased gradually for 48h, the best dose was 1-5mM. 1-5mM Shuanghuanglian had protective effect on acetaminophen injuried Canine hepatocytes.
