Abstract:
To explore the application of orthogonal design in screening combination of straw-degrading bacteria, optimize combination of straw-degrading bacteria producing cellulase and ligninase. With corn straw as a fermentation substrate, the straw-degrading rate and enzyme activity of different combinations of degrading bacteria were investigated with orthogonal design. The antagonism between strains in selected combinations was studied with plate test. Results showed that, the difference of straw-degrading rates among different treatments was extremely significant. The optimal combination was A1B3C1D4E1F2G1, i.e., the inoculation volume of stain F-11, XX-10 and G-09 was 1 mL, 2.0 mL and 1.0 mL, respectively. The degradation rate increased by 10% compared with that of the control. The antagonism existed between some straw-degrading bacteria, and the straw-degrading rate was related to strain composition in combination. The orthogonal design was an effective method to screen the best combination of straw-degrading bacteria, and the selected bacteria combination had high degrading efficiency for straw and very good value of development and utilization.
