In Vitro and in Vivo Antioxidant Effects and Mechanism of Selenium –enriched and Germanium –enriched Inonotus obliquus polyphenol

Abstract:

The aim was to investigate the antioxidant effects of Inonotus obliquus polyphenol (IOP), Selenium-enriched Inonotus obliquus polyphenol (Se-IOP) and Germanium-enriched Inonotus obliquus polyphenol (Ge-IOP). The reagent kit methods were used to determine anti-reactive oxygen species (anti-ROS) unit and malondialdehyde (MDA) value, the spectrophotometry methods were used to determine lipoxygenase activity, inhibitory rate of red blood cell (RBC) hemolysis and glutathione peroxidase (GSH-Px) activity, respectively. In chemical modified systems, the antioxidant activity of 4.00 mg/mL IOP, Se-IOP and Ge-IOP were 52.36, 89.21 and 82.93 anti-ROS units, the inhibiting rates on lipoxygenase were 34.15%, 42.22% and 50.53%, respectively. In vitro cellular and tissular antioxidant systems, the inhibitory rate of 4.00 mg/mL IOP on mice RBC hemolysis by H2O2 inducement was 69.65%, which of 1-4.0 mg/mL Se-IOP and Ge-IOP were above 100%. The inhibitory effects of 1-4.0 mg/mL IOP, Se-IOP and Ge-IOP on the formation of MDA in mice without inducement were very significantly as compared with control group (P<0.01). In vivo antioxiddant systems, the MDA value in mice liver decreased very significantly when the mice were intra-peritoneal injected with IOP, Se-IOP and Ge-IOP (4.0 mg/mL,0.5 mL/d, 12 d) respectively(P<0.01), the GSH-Px activities in mice serum and liver also increased very significantly after being injected with Se-IOP and Ge-IOP(1-4.0 mg/mL,0.5 mL/d, 12 d) respectively (P<0.01). IOP, Se-IOP and Ge-IOP had antioxidant effect in vitro and in vivo, Selenium and Germanium could increase the antioxidant activity of Inonotus obliquus polyphenol.
