Abstract:
Nine Panax ginseng genus squalene synthase in Genebank was analyzed in this article. Squalene synthase cDNA sequence and its encoded amino acid sequence, physicochemical properties, the signal peptide, hydrophobic, hydrophilic, subcellular localization, transmembrane topological structure, function domain and evolutional relationship were predicted by using bioinformatics methods. The result showed the similarity of ginseng squalene synthase nucleotide was 96.245%, and the sequence of amino acid shared 95.5% similarity on average. Furthermore, the main composition of 9 amino acid sequences of squalene synthase were alpha helix and random coil via secondary structure predicted. Nine ginseng species could be divided into two subtribes by phylogenetic tree analysis. The phylogenetic tree which included nine ginseng plants and other species of plants, animals, yeast showed they belong to different groups. By analyzing the biological characteristics of ginseng squalene synthase, it could provide theoretical reference for the squalene synthase gene cloning and genetic manipulation.
