Abstract:
With the improvement of people’s requirements of the agricultural product quality and safety, efficient screening of biological control of resources becomes particularly important. A Bacillus, numbered YW26, inhibiting a variety of plant pathogenic fungi was isolated and identified by plate confrontation experiments, molecular identification, Congo red staining, cloning of glucanase gene and its sequence analysis in this study. 16S rDNA gene sequence analysis showed that it was a strain of Bacillus amyloliquefaciens. The strain had a strong cellulase production capacity by Congo red staining analysis. Primers were designed according to Bacillus endoglucanase gene (CP000560) in the GenBank database, a 1527 bp fragment was cloned, and the fragment had 99% homology with endo-β-1,4-glucanase gene sequence of Bacillus amyloliquefaciens FZB42 by sequencing and BLAST analysis. The Bacillus amyloliquefaciens strain had significant effect of inhibiting pathogens as well as strong cellulase production capacity. Therefore, the strain can be used as biological control resources to develop new biological control agents.
