Cloning and Induced Expression Analysis of tps1 Gene of Fusarium Oxysporum f. sp.

Abstract:

In order to realize the difference of tps1 gene between Fusarium oxysporum f. sp. FOC race 1 and race 4 and other Fusarium oxysporum f. sp. and analyze whether the host selective difference between FOC race 1 and race 4 were related with tps1 gene. The tps1 genes of race 1 and 4 were amplified by Polymerase Chain Reaction and Reverse Transcription Polymerase Chain Reaction and sequence, the gene sequences and the encoded protein sequences were compared by DNA Star software, and also analyzed the expression quantity alteration of tps1 after induction with Musa AAA Cavendish or Musa ABB Pisangawak. The full length of tps1 from FOC race 1 and race 4 were both 1660 bp, with their open reading frames 1605 bp, encoding 535 amino acids. There were only two base differences among the sequences of the tps1 gene of FOC race 1 and race 4 and their amino acid sequences were the same. The tps1 gene sequences of these two races were different from those of other specialized forms or species of Fusarium oxysporum f. sp. It was inferred from the differences of the tps1 gene sequence and protein sequences from FOC1 and FOC4 isolates that there was no obvious relation between tps1 gene and the host selective preference of FOC1 and FOC4. These results would help to know the relation of tps1 genes and FOC long-term survival without host in the future research.
