In silico cloning and sequence analysis of glucanase gene(ScBG1) from sugarcane

Abstract: In order to reveal the structure and function characteristics of β-1,3-glucanase gene in sugarcane, and provide the basis for the research and application of this gene, a sugarcane full-length cDNA sequence of β-1,3-glucanase gene, termed as ScBG1, was obtained in silico by using the rice β-1,3- glucanase gene (U72255.1) as search probe, and the bioinformatics analysis was also finished. The results showed that the full-length of the cDNA sequence of the ScBG1 gene was 1842 bp, which contained a 1488 bp complete open reading frame and enco ded 495 amino acid residues. The assemble gene belonged to the 17th glycol hydro family and hydrophobic protein containing the signal peptide. With several plants’ β-1,3-glucanase Based on the sequences comparison of the ScBG1 gene with β-1,3-glucanase genes from several other plant species, the ScBG1 gene had the typical structural region of β-1,3-glucanase and was high conserved with 45%-73% sequence similarity. The results in this paper provided the basis for experimental isolation of the ScBG1 gene in sugarcane and the reference for the research of its molecule function.
