Adventitious Buds Induction and Elongation from Mature Zygotic Embryo of Larix principis-rupprechtii Mayr.

Abstract: In this study, mature zygotic embryos of Larix principis-rupprechtii Mayr. was used for a preliminary investigation on the organogenesis of Larix principis-rupprechtii Mayr.. Different disinfection methods were utilized to process with the explants and an orthogonal experiment designed with 6-BA, NAA and sucrose was employed to investigate the induction of adventitious buds. Moreover, a few activated carbons were added to the medium in the elongation phase. The results showed that (1) the optimal disinfection method was that after rinsed in running water for 12 hours, the seeds were surface decontaminated with 75% ethanol for 3 minutes and then disinfected with 0.1% mercuric chloride solution for 10 minutes; (2) The best combination of adventitious buds induction was DCR medium supplemented with 4 mg/L 6-BA, 0.1 mg/L NAA and 40 g/L sucrose under a light intensity of 2000 lx, in which NAA and sucrose were no significant effect; (3) 1.0 g/L activated carbon played a positive role in elongating of adventitious buds, which was 0.43 cm average elongation. It is significantly theoretical and practical basis for the organogenesis and further genetic transformation research of Larix principis-rupprechtii Mayr..
