Adventitious Bud Regeneration from Leaves of Loropetalum chinense var. rubrum Cultivars

Abstract: The aims were to establish Loropetalum chinense var. rubrum cultivars efficient regeneration system and lay a foundation for solving the problem of gene engineering breeding. In this paper, the regeneration system of Loropetalum chinense var. rubrum cultivars in vitro was studied with leaves as explants. Some factors such as different cultivars, illumination condition, and hormone concentration and so on were investigated. The results showed the regeneration efficiency of leaves was different. The highest one was in L. chinense var. rubrum‘MeihongXiyeqing’, followed L. chinense var. rubrum‘Changye Meihong’and variegated variation of L. chinense var. rubrum No.2. It could have higher regeneration efficiency when the leaves with the treatment by white light and red light and laid on the medium in adaxial side down, after 70 days, their regeneration frequency were 50% and 25%. The optimal bud regeneration medium from leaves of L. chinense var. rubrum‘Meihong Xiyeqing’was MS + 6- BA 1.0 mg/L + NAA 0.5 mg/L, the induction frequency was 41.6% . The appropriate medium for adventitious bud induction from leaves of L. chinense var. rubrum‘Changye Meihong’was MS+6-BA1.0 mg/L+NAA1.0 mg/L, the induction frequency was 33.3%.
