Optimization of Anticoagulant Substance Extraction Methods of Poecilobdella manillensis

Abstract: In order to select the optimal extraction method of anticoagulant substance of Poecilobdella manillensis, 12 kinds of extraction methods were used to extract anticoagulant substance from living (6 kinds) and dry (6 kinds) body of P. manillensis, respectively, and three kinds of extraction methods were used to extract anticoagulant substance from salivary gland secretions of P. manillensis. The results showed that 80% pre-cooled acetone were the best for extracting anticoagulant substance from living and dry body of P. manillensis, and the amount of extraction was up to (61.20±1.13) AT-U/g and (89.19±1.13) AT-U/g, respectively. As to salivary gland secretions of P. manillensis, the amount of anti-coagulant substance was up to (24.41±0.98) AT-U/g using 10% trichloroacetic acid. The research provided optimal extraction method, which was simple and practicable, of anticoagulant substance from living and dry body and salivary gland secretions of P. manillensis, and would contribute to large-scale production of hirudin.
