Key Techniques for Rapid Propagation of Haworthia cooperi var. pilfera of Liliaceae

Abstract: The research aims to establish a rapid regeneration system of Haworthia cooperi var. pilfera. The authors used the inflorescence as explants and MS as the basic medium to research the effect of different proportions of hormones on inducing differentiation, growth, expanding propagation, strong young plants and producing roots. The efficient technique system for rapid propagation was established. The result showed that the optimal priming medium was MS 6-BA 2.0 mg/L NAA 0.02 mg/L and the optimal induction and differentiation medium was MS 6-BA 0.5 mg/L KT 0.5 mg/L NAA 0.01 mg/L and the callus induction rate was 87.1%, the bud differentiation rate reached 52.3%. The best rooting medium was 1/2MS IBA 0.5 mg/L NAA 0.1 mg/L AC 0.5 g/L. The root growing rate of tube-seeding was 97.5% and the transplanted seedling had a survival rate up to 90%. The techniques based on this research for tissue culture and rapid propagation had no damage to the female and could be applied to large-scale production of Haworthia cooperi var. pilfera seedlings.
