Study on Effects of in vitro Dormant Branches of Trees Garden Material of Osmotic in Artificial Lasting Low Temperature

Abstract:

To study the changes of substance of the branches of trees within the garden in vitro permeability under the stress of continues low temperature -10℃,the essay was to provide a theoretical basis for the deep sleep physiological changes in vitro the branches. Experiment uses Prunus×Cistena pissardii, Cotinus coggygria ‘Atropurpureus’, Acer palmatum, Acer truncatum in the deep dormancy for the tested material. Branches in water in vitro conditions, experience -10℃ low-temperature stress provided by refrigerator, which repeat 3 times. The results showed that: Prunus×Cistena pissardii branches of soluble proteins showed decreased changes, Cotinus coggygria ‘Atropurpureus’ first increased then decreased, and Acer palmatum and Acer truncatum showing the change has been rising; after 3-12 days, Cotinus coggygria ‘Atropurpureus’ proline content decreased gradually, gradual increased in the other 3 trees; after 12-15 days, Prunus×Cistena pissardii and Acer palmatum declined slightly, the other two trees showed up changes; 4 garden trees soluble sugar content showed rising change in vitro shoots, the 15th to the highest value. Comprehensive analysis believed that when branches of the trees were in deep dormancy, osmotica changes under low temperature stress appear certain physiological activity, and when they were under -10℃ low temperature stress in the range of 3-9 days, permeability changes in the material seemed no significant difference in levels.
