The Effect of Organic Compounds on the Propagation of Phalaenosis PLB and Anatomical Observation

Abstract:

The Phalaenopsis is a kind of expensive flower and its needt is large in the marke, the tissue culture of the Phalaenopsis can meet its great need of the market. The Phalaenopsis PLB as explants were inoculated on the improved VW + 3.0 mg/L 6-BA + 0.2 mg/L NAA + 20 g/L surose + 16 g/L agar medium + 1 g/L active carbon adding five different organic compounds (coconut milk, coconut juice, banana mud, potato, casein hydrollysate) with different concentrations (10%,15%,20%). The results showed that five organic compounds which had effect on PLB propagation from high to low were coconut milk, coconut juice, potato, banana mud and casein hydrollysate, respectively. The best concentration of coconut milk for propagation was 15%, coconut juice was 20%, potato juice was 10%, banana mud was 15%, but 10%, 15%, 20% were so high to casein hydrolyzed, so restrained the propagation of Phalaenosis PLB. Primary protocorm-like body (PLB), secondary PLB and leaf bud induced by PLB of Phalaenopsis were observed by free-hand section method, and their connection and differentiation were analyzed. The results showed that the inner cells in primary PLB were big and irregular and the cells in secondary PLB were compacted and homogeneous. Secondary PLB were formed from the hyposphere of primary PLB, and leaf bud was formed from the episphere of primary PLB. Semi-moon-shaped stoma and “multi-rod structure” were found in primary PLB, and “multi-rod structure” function was unknown.
