Analysis on the Genetic Stability of Agaricus bisporus Strains Preserved in Liquid Nitrogen

Abstract:

After four years’ preservation in liquid nitrogen, four Agaricus bisporus strains, i.e. As2796, As3003, As4607 and As4580, were thawed and detected for the survival ratio. The survived cultures were put into mycelia running test in compost with different water content, different pH value and under different temperature. Moreover, their cellulase activity, isozyme patterns, RAPD patterns and fruiting ability were also checked. The experimental results showed that after four years’ liquid nitrogen preservation, the stock cultures of A. bisporus had the same mycilia growth speed and cellulase activity as the control, and they kept the same isozyme and RAPD patterns, as well as the original fruit-bodies growing characteristics, production and quality. Therefore, the liquid nitrogen preservation could keep the stability of A. bisporus stock culture in a long time.
