Effects of Color Plastic Films on Induction of Callus of Stems and Three Main Alkaloids Contents in Phellodendron amurense

Abstract:

The stems of Phellodendron amurense were used as explants to describe the effects of different color plastic films on induction of callus and on the contents of berberine, jatrorrhizine, palmatine. The stems which were inoculated on MS medium containing 1.5 mg/L 6-BA, 0.5 mg/L NAA and 20 g/L glucose were treated under different color plastic films. After 20 days of treatment, morphological characteristics of callus were observed and biomass and alkaloid contents were determined. It was shown that all the four treatments induced callus of stems and the induction rate was 100%, but the effects on the status and growth of callus and content of secondary metabolic product were different under various color plastic films. The produced callus tissues under control and blue film were vivid, loose in texture, fast in growth while fewer and aged callus tissues were induced under other films. Further studies showed that the contents of the three alkaloids under control and blue film were higher than others. The results showed that control and blue treatment were the best for growth of tissue and content of secondary metabolic product, followed by yellow and the worst were under green and red ones.
