Establishment of CAPS Molecular Marker for Ty-1 Gene Resistant to Tomato Yellow Leaf Curl Disease

Abstract:

The study was designed to develop and obtain molecular markers tightly linked to Ty-1 gene resistant to tomato yellow leaf curl virus, in order to affording technical supports for tomato disease resistance breeding. In this study, resistant heterozygotes (Ty-1/ty-1), resistant homozygotes (Ty-1/Ty-1) and susceptible homozygotes (ty-1/ty-1) were used as experimental materials. Based on the published RFLP markers tightly linked to Ty-1, four pairs of specific primers were designed for PCR amplification. PCR products were then digested with different restriction enzymes to develop CAPS molecular markers. As a result, two steady-going and available CAPS molecular markers were found, i.e. TG97 and Mi23. TG97 marker showed three restriction fragments (398 bp, 303 bp and 95 bp) with resistant heterozygous materials, two restriction fragments (303 bp and 95 bp) with resistant homozygous materials and only 398 bp fragment with susceptible homozygous materials. Mi23 marker showed two restriction fragments (402 bp and 354 bp) with resistant heterozygous materials, only 402 bp fragments with resistant homozygous materials and only 354 bp fragments with susceptible homozygous materials. These findings indicated that the TG97 and Mi23 were co-dominant CAPS markers, which could be used in tomato breeding for disease resistance.
