Creation and Quality Analysis of Glu-D1 Subunit Absence and Weak Gluten Bread Wheat (Triticum aestivum L.)

Abstract:

In order to obtain diversity of wheat quality traits, the cell lines derived from immature embryos of bread wheat were treated with 60Co-? radioactivity. The plants without High-Molecular-Weight glutenin subunit (HMW-GS) controlled by locus Glu-D1 were found from regeneration offsprings of cultivar ‘Ji3235’. By further selection, seven heredity-stable variation lines were got. There was less difference in agronomy traits and gliadin electrophoresis types between the seven lines and ‘Ji3235’. However, the obvious variations were that the seven lines did not have HMW-GS of Glu-D1 in SDS-PAGE (Ji3235 contained subunits 2 + 12) and their flour quality were very poor, namely the wet gluten could not be detected and the sedimentation values became very low. These variation lines are important materials for breeding weak gluten wheat cultivars and evaluating the roles of Glu-D1 locus at wheat quality.
