Purity Detection and Fingerprint Construction of ‘Xiangnongdamian 1’ Hybrid with SSR Markers

Abstract:

In order to identify the truth and purity of hybrid seeds rapidly and accurately, thirty three pairs of SSR primers were selected to detect the polymorphism of ‘Xiangnongdamian 1’ and its parents according to the origin of the parent materials and the proposed SSR core primer pairs. The results showed that: eight pair primers were amplified with clear and stable fragments among the parents and their hybrid, and the all polymorphic sites were codominant markers. All the codominant markers can be used to identify the purity of hybrid mixed with parents and other seeds during seed production. Digital fingerprint of ‘Xiangnongdamian 1’ and its parents were constructed with the eight primers amplification results by changing binary system into decimal system, which can provide much more accurate technological instructions for truth and purity detection of hybrid and parent materials.
