Abstract:
To widen the explants source and promote the development of tissue culture technology of Hevea brasiliensis. The leaves from sterile seedlings of rubber tree as explants were induced to calla on media containing different concentrations and combinations of plant hormones. When the concentration of 6-BA was definite, the induction rate of callus was gradually improved with the increasing of NAA concentration. The rate was the highest, while the concentration of 6-BA reached 1.5 mg/L, and more than 1.5 mg/L, the rate was gradually declining. The results of different concentrations and combinations of KT and NAA showed that the calla induction rates of most media were less than ten percent, some were even zero. When NAA had the same concentration, the ability of callus induction from 6-BA was obviously superior to KT. The callus induction rate of leaf must depend on plant hormone in Hevea brasiliensis. The calla can not be induced when the media had no hormone or containing single hormone. The modified MS+1.5 mg/L NAA+1.0 mg/L 6-BA was the most appropriate medium of callus induction rate from leaf, and the rate reached to 26.7%.
