Varieties Identification of Tomato Using Molecular Markers

Abstract: Three PCR-based systems, including RAPD, RGA and SRAP were employed to differentiate 22 tomato varieties. In this study, laborious work was conducted in primer selection, 17 polymorphic loci were produced by 7 RAPD primers, 12 polymorphic loci by 2 RGA primer pairs, and 8 polymorphic loci by one SRAP primer pairs. The informative loci by each primer or primer pair were used to identify 22 tomato varieties, respectively. It showed that the RGA and SRAP marker were more effective. To distinguish the 10 varieties, two-primer-combinations and three-primer-combinations were tested. Three-primer-combination BI26、PK1+PK2 and S91 or S92 can identify the 22 tomato varieties effectively and produces specific fingerprint of each varieties. More over, cluster analyses indicated that the relationship between varieties was very complicated, and the cluster analysis could not get the desired result with limited informative data.
