Preliminary Comparative Proteomics of Young and Mature Leaves of Hevea brasiliensis

Abstract:

In order to demonstrate the metabolism functions in leaves during different development stages and reveal the control mechanism of natural rubber biosynthesis in latex in Hevea brasiliensis, the bronze period of young leaves and the stabilization stage of fully-expanded mature leaves from H. brasiliensis were used as materials to perform proteomic analysis in this study. The leaf total proteins were extracted and then separated by 2-DE. Comparison of the 2-DE gels for the young and mature leaves revealed many different expression protein spots, and most of these spots were finally identified by MS as carbon metabolism enzymes and posttranslational modification related proteins and so on. Conclusion: A mature protein extraction and 2-DE system for H. brasiliensis leaves was established, and a high resolution 2-DE reference map for leaves was obtained. Most different expression proteins such as hydroxynitrile lyase, Rubisco and actin were identified in young leaves and/or mature leaves. These results may has important roles for revealing natural rubber biosynthesis mechanism and provide some reference data for proteomic analysis of other tropical plants.
