Germplasm Resources Analysis in Cyprinus carpio var. jian by Microsatellite Markers

Abstract:

For evaluating and rational utilization the germplasm resources of Cyprinus carpio var. jian. 185 individuals were randomly sampled from breeding population of C. carpio var. jian, and genetic diversity were analyzed using 20 microsatellite markers. The results showed that the DNA fragment length of the alleles ranged from 124-310 bp, a total of 156 alleles and 172 genotypes were detected in the population by 20 microsatellite loci, the number of allele and genotype at each locus ranged from 5 to 13 and from 10 to 44,with the average of 7.8 and 20.1 respectively. The observed and expected heterozygosity varied from 0.346 to 0.978 and from 0.619 to 0.880, with the average 0.6434 and 0.757 respectively. The polymorphic information content (PIC) was 0.552 to 0.868, and the average was 0.7253, all the loci were in a high polymorphism degree (PIC>0.5). The results indicated rich polymorphism information content and large genetic diversity in the population, and this introduced the breeding population of C. carpio var. jian has a large selection potential. The average fixation index (FIX) was 0.1479 indicated that heterozygote deficiency existed in the population. The UPGAM dendrogram based on the genetic distance could clearly showed the genetic diversity of all the individuals, and this could provided the basis for breed conservation, chosen breeding pairs and avoid in breeding.
