Abstract:
The aims were to screen and identify a microorganism strain with absorbing and resisting to copper and to use it removing heavy metal copper in the environment. By using microorganism screening and separating methods, the copper-resisting microorganism were widely obtained from the soil around pig farms with a long captive time. The target microorganism strains were screened and purified repeatedly. Their morphology and molecular identification were analyzed and the microorganism strains were applied to absorb heavy metal copper in tea garden soil. Morphological characteristics were observed under the 400 times optical microscope and scanning electron microscope. Amplification of DNA sequencing fragment based on the 5.8S rDNA universal primers were compared in the GenBank database using the BLAST tool. 10 strains of fungus of different copper-resisting concentrations were separated. The highest concentration of copper-resisting strain of the fungus was A4 which concentration of copper-resisting reaches 940 mg/L. A4 bacteria grew fast in Czapek medium. It was identified synthetically as Aspergillus fumigatus. The strain A4 had good absorption to copper in tea garden soil.
