Studies on Pollen Germination and Storage of Iris ensata Thunb.

Abstract:

The effect of different media on pollen germination of Iris ensata through orthogonal experiment, in other hand, the effects of storage condition, storage temperature and storage time on pollen germination were studied. The results showed that: (1)The optimum medium was sucrose 200 g/L +H3BO3 300 mg/L +CaCl2 200-300 mg/L, in which the rate of germination was 81.55%. (2) Dry conditions was suitable for pollen storage of Iris ensata, and the silica gel was the suitable desiccant. (3)The pollen viability decreased over time, but the viability decreased slowly with the falling of temperature, pollen stored at -196℃ for 270 days, its germination rate was 84.21%, which was not significant with fresh pollen.
