Genetic Diversity Analysis of Cauliflower Germplasm by SRAP Markers

Abstract: In order to reveal the genetic relationship of cauliflower germplasm at molecular level, providing some theoretical basis for germplasm collection, identification, utilization, genetic improvement. The genetic diversity of 24 genotypes of cauliflower (Brassica oleracea L. var. botrytis L.) germplasm was evaluated by sequence- related amplified polymorphism (SRAP) technology in this study. A total of 257 alleles were detected by 23 SRAP markers,which screened from 30 primer combinations. The average bands of each primers combination ranged from 5 to 20, of which 185 ones were polymorphic and the polymorphism frequency was 71.98%, indicating that the cauliflower germplasm relatively had extensive genetic diversity. Similarity coefficient analysis showed that genetic similarity among genotypes was from 0.5491 to 0.9626, averaged out at 0.7490. Cluster analysis showed that origin and district could be used as agronomic traits in cauliflower germplasm cluster analysis.
