Callus Tissue Culture in Dioscorea nipponica Mak

Abstract: The callus culture on Dioscorea nipponica(leafstalk, stem, leaf, stem tip) was conducted. The results show that the calli induced from different parts have capability to produce Diosgenin which is the secondary metabolism. The stem tip is better than other parts in callus growth and Diosgenin content is influenced by some culture conditions, such as media, PH, temperature and hormone. The optimum cultural conditions on callus culture is incubated in medium with modified medium and 2,4-D(3.0~5.0mg/L) and BA(1.0mg/L),which enables the content of Diosgenin highly and the callus growth quickly.
