Study on Grain Formation Physiology of TGMS Rice with eui Gene During Grain Filling Period

Abstract: In order to investigate the grain formation physiology of thermo-sensitive genic male sterile (TGMS) rice with the elongated uppermost internode (eui) gene during grain filling period. TGMS rice ‘Changxuan 3S’, possessing eui gene, was studied by using low temperature underground water to irrigate TGMS rice line ‘Changxuan 3S’ to multiply it, with its parent ‘Pei’ai 64S’ used as a control. The results showed that, With the process of grain filling, the contents of amylase and amylopectin increased quickly at 7-27 days after flowering, then increased slowly and stopped increasing at 31 days after flowering, and the soluble protein content and the activities of the ADPG pyrophosphorylase (ADPG-PPase), glutamine synthetase (GS) and glutamate synthase (GOGAT) decreased gradually, and the activities of the soluble starch synthase (SSS) and starch branching enzyme (SBE) increased gradually and then declined slowly, showing a single-peak curve in the grains of ‘Changxuan 3S’ and ‘Pei’ai 64S’. Throughout the filling stage, the contents of amylase, amylopectin and soluble protein and the activities of ADPG-PPase, SSS, SBE, GS, GOGAT in the grains of ‘Changxuan 3S’ were higher than those of ‘Pei’ai 64S’, but its bagged seed set and per plant yield were lower than those of ‘Pei’ai 64S’. Differences analysis showed that the differences of the 5 enzymes activities, amylase content, amylopectin content, bagged seed set and per plant yield were not obvious, and the difference of soluble protein content was obvious in filling grains of ‘Changxuan 3S’ and ‘Pei’ai 64S’.
