The Study of Efficient Plant Regeneration of Lycopersivon esculentum Mill.‘Huangyingtao No 2'

Abstract: The effects of different hormone combinations on callus induction and bud differentiation of cotyledons in Lycopersivon esculentum Mill.‘Huangyingtao No 2’were studied. Differences in numbers of callus and regeneration buds were showed under different hormone treatments. The optimum medium to induce callus and regeneration buds was MS+ 6-BA 2.5 mg/L+ IAA 0.2 mg/L, with induction of callus rate up to 95.08% and regeneration buds averaged five. Roots of generation buds were induced normally in 1/2MS+ IAA 0.2 mg/L. According to effects of cefotaxime (Cef) and carbenicillin (Carb) to cotyledons regeneration, medium to remove agrobacterium was MS+ 6-BA 2.5 mg/L+ IAA 0.2 mg/L+ Cef 200 mg/L+ Carb 200 mg/L. Meanwhile, as a widely used selective agent for transgenic plants, kanamycin should be applied at concentration which can not only select positive plants efficiently but also do no harm to cotyledons regeneration. After treatments of kanamycin (Kan) to cotyledons at different concentration, 50 mg/L was found to be the optimum concentration.
