Primary Studies on the Induction and Growth of Callus of Sinojackia xylocarpa Hu

Abstract: In order to primary studies on the collection time of explants and induction and growth of callus of tender leaf (Sinojackia xylocarpa Hu), the leaves used as explants with WPM+2,4-D 2.0 mg/L+6-BA 1.0 mg/L+ sucrose 20 g/L+ agar 8 g/L as medium for the experiment to study collection and disinfected time on tender tissue culture. The results showed that the optimal time of explants collection for tender tissue culture was April. During this period, the contamination rate of explants was the lowest. The optimal sterilizing treatment for the tender leaves was 75% Alcohol 30 s+0.1% HgC12 1-5 min. Different disinfected time had a distinctly effect on physiology and biochemistry of callus. The soluble sugar in callus inducted with 1 min and 3 min disinfected time were significantly higher (P<0.05) compared with 5 min disinfected time. The soluble protein in callus inducted with 1 min and 3 min disinfected time was significantly lower (P<0.05) than that in callus with 5 min disinfected time. Different sterilization time had no significantly effect on the activity of peroxidase and superoxide dismutase. The callus inducted with 3 min and 5 min disinfected time had a stronger tolerance (P<0.05) of salt stress compared with the callus inducted with 1 min disinfected time.
