Establishment of Leaf-explant Regeneration System in Vitro of Triploid Hybrids of White Poplar ‘Beilixiongzhu 1’and ‘Beilinxiongzhu 2’

Abstract: Abastract: To solve the difficulties in vegetative propagation of triploid hybrids of white poplar '‘Beilinxiongzhu 1’and ‘Beilinxiongzhu 2’, the effects on disinfection method and adventitious buds from different hormones combination were studied by taking semi-woody shoots came from triploid hybrids of white poplar ‘Beilinxiongzhu 1’and ‘Beilinxiongzhu 2’ as explants. The results indicated that: (1) the suitable method for disinfection of the two triploid hybrids was to treat for 20 minutes by 20% 84 disinfectant. (2)6-BA and TDZ had significant effects on regeneration rate and coefficient of the adventitious buds. However, NAA had insignificant effect on regeneration rate of the adventitious buds, but significant effect on coefficient of the adventitious buds. The optimum medium for the regeneration of adventitious bud from leaves in ‘Beilinxiongzhu 1’ was MS+ 0.2 mg/L 6-BA+ 0.1 mg/L NAA+ 0.005 mg/L TDZ. (3) Significant effects on regeneration rate and coefficient of the adventitious buds of 6-BA, NAA, and TDZ were observed in the triploid hybrid ‘Beilinxiongzhu 2’. The optimum medium for effective inducing adventitious buds in vitro leaves was MS+ 1.0 mg/L 6-BA+ 0.05 mg/L NAA+ 1.2 mg/L ZT. The rapid propagation systems of leaf explants in the two triploid hybrids were successfully established, which provided a platform for large-scale vegetative propagation and further genetic transformation of ‘Beilinxiongzhu 1’ and ‘Beilinxiongzhu 2’.
