Abstract: The purpose of this study was to identify the salt tolerance of different cotton varieties in seedling stage. Solution culture experiments were carried out to study the effects of 0.3% NaCl stress treatment for 24 hours, 48 hours and 72 hours on the physiological and ecological indexes of six cotton varieties’ seedling, including lipid peroxidation, membrane integrity, reactive oxygen species (ROS) accumulation and antioxidant enzyme activity of roots. The results showed that there was no significant difference of membrane integrity and ROS accumulation in seedlings treated with NaCl for 24 hours. NaCl stress treatment for 48-72 hours significantly accelerated ROS accumulation, increased membrane lipid peroxidation and destroyed membrane integrity of seedlings. The malondialdehyde accumulation and membrane relative permeability increase were proved by Schiff′s reagent and Evans blue. The synthetic assessment of the six parameters indicated that under 0.3% NaCl stress for 72 hours, the order of salt tolerance of different cotton varieties was ‘N181’>‘N52’>‘N177’>‘K-1’>‘N203’>‘Zhong50’.
