The Clone and Sequence Analysis of BiCDPK Conserved Sequence from Bothriochloa ischaemum

Abstract: In order to study the structure and function of CDPK gene, BiCDPK was cloned from Bothriochloa ischaemum by using RT-PCR method, and analyzed by bioinformatics. The results showed that the length of cDNA for BiCDPK was 1048 bp, which contained a 387 bp complete open reading frame encoding 128 amino acids. The corresponding protein was mildly acidic, stable and hydrophilic, with random coil and α-helix as main components in secondary structure. Subcellular localization indicated that the protein was located in cytoplasmic without transmembrane region and signal peptide. The sequence of protein included one serine phosphorylation sites and three threonine phosphorylation sites. Further phylogenic analysis displayed that BiCDPK in Zea mays, Triticum aestivum and Oryza sativa shared 97%, 86% and 86% identity. It laid a foundation to further study the BiCDPK function in Bothriochloa ischaemum.
