Agrobacterium tumefaciens-mediated Transformation of Rice for gna Gene

Abstract: Brown plant hopper distributes very wide, and it has been the major rice insects in Southern China. Many studies showed that it was economical and effective method to control insects with transgenic breeding. An expression vector was constructed with a safe marker gene, 6-phosphomannose isomerase (PMI), and an insect-resistant gene, Galanthus nivalis agglutinin(GNA), was used in rice genetic transformation in the present study. Results indicated that the pmi and gna gene have been expressed and transferred into some rice cultivars with PCR analysis, southern blotting and GUS detection, and the gna gene was mainly integrated into rice genome in manner of single copy. Some mature seeds randomly selected from eight transgenic rice plants were cultured 15 days on 1/2 MS medium with 50 mg/L hygromycin, and numbered the resistant and sensitive seedlings. The results indicated that most of the transgenic plants were according with the law of single gene segregation.
