RAPD Analysis of the Amorphophallus albus Germplasm

Abstract: Amorphophallus albus native to China is one of the precious wild species in the genus Amorphophallus. The rich genetic variations were also found within the species. Using the RAPD techniques, the phylogenetic relationships among the different bio-types or origin places of the species were analyzed in the present study. The genomic DNAs isolated from 34 accessions of A. albus were amplified by using 40 decamer primers. Eleven primers showed consistent banding patterns and amplifications in all the materials tested and produced 112 bands, of which 93 bonds were polymorphic. Cluster analysis using dps program shows that the 43 materials were divided into 5 groups. A comparison of this result with the leaf-stem morphology, we found that the clusters were highly correlated with the geographic origins of the materials. This result provided molecular information for a better understanding of the genetic relationship of A.albus species.
