Assessment of Genetic Diversity of Maize Detected by RAPD Molecular Markers

Abstract: In this study, RAPD molecular markers were used with PCR in DNA of 10 maize individuals, then the results of PCR were analyzed by Popgene32 and SPSS 13.0, to assess genetic diversity among ten maize individuals. The results showed that: (1) 214 RAPD bands were amplified by 20 RAPD random primers with PCR; (2) the percentages of polymorphic loci of the screened primers are 86.4%, the genetic similarity of among ten maize varieties is from 0.316 to 0.654; (3) Jaccard’s genetic similarity matrix and dendrogram for ten kinds of maize were formed. In these dendrogram, ten individuals could be divided into three groups; (4) RAPD molecular markers are suitable for DNA fingerprinting, identification and genetic analysis in maize.
