Study on Progenitive Technology of Test-tube Micro- bulbodium of Amorphophallus konjac

Abstract: The question of the low propagation coefficient in the production of Amorphophallus at present, the research may use in the large scale production induction technology for test-tube micro-bulbodium. This experiment cut to the seedling for callus differentiation through three different methods, separately setting in the different hormone allocated proportion four kind of culture medium. Results showed that callus was induced and differentiated in media consisted of MS+ 6-BA2.0 mg/L +NAA0.5 mg/L + 6% sucrose and MS+ 6-BA0.8mg/L +NAA0.5 mg/L + 6% sucrose. 1/2MS+ NAA0.08 mg/L + 6% sucrose and 1/2MS+ IBA0.8 mg/L + 6% sucrose were fit for the base of the stem with some callus for rooting, and 1/2MS+ NAA0.08 mg/L + 6% sucrose was suitable for the base of the stem with little callus to induce micro-bulbodium, breaking from the base of stem , there is the bud spot as the micro-bulbodium. It was discovered that the formation of test-tube Micro- bulbodium not only with the culture medium composition related, but also with the way of cutting for the seedling base
