Cloning and functional analysis of the promoter of small rubber particle protein (SRPP) gene in Hevea brasiliensis

Abstract: Abstact: The small rubber particle protein (SRPP) is an essential factor related to rubber biosynthesis in Hevea brasiliensis (rubber tree). The amino acid sequence of SRPP has high similarity to those of rubber elongation factor (REF) and phaseolus vulgaris stress-related protein (PvSRP), which is 72% and 68% respectively. For further investigation of the expression of SRPP gene, the promoter of SRPP gene was cloned by the method of ligation-mediated PCR. Sequence analysis demonstrated that light-responsive elements accounted for 39% in the promoter of SRPP gene, which indicates that SRPP promoter might be light inducible and that the expression of SRPP gene could be regulated by light. In addition, the promoter of SRPP gene contains a variety of stress-responsive elements, such as HSE,MBS, TC-rich repeats, ABRE, EIRE and W box etc. These data suggest that SRPP is not only involved in rubber biosynthesis, but also plays an important role in response to various stresses in rubber trees.
