Preliminary optimization the extration conditions of triterpene in callus of Betula platyphylla Suk.

Abstract: (OBJECTIVE)To improve the yield of triterpene in callus of birch, (METHOD)oleanolic acid as standard, 5% vanillin-acetic acid and perchloric acid as the developer, the determination method of triterpene was evaluated, the results showed that this method had a higher repeatability, and quite stable during 5~50min in color reaction; The triterpene in callus with ultrasonic was extracted under 20℃,40℃,60℃,80℃, the content of triterpene was 40℃(1.67mg/g)> 60℃(1.28 mg/g)> 80℃(0.47 mg/g)> 20℃(0.08 mg/g). The triterpene in callus was extracted with benzene, acetone, ether, ethanol95%, methanol, chloroform, the content of triterpene was benzene(2.58 mg/g)> ether(2.04 mg/g)> ethanol(1.67 mg/g)> chloroform(1.21 mg/g)> methanol(0.89 mg/g)> acetone(0.50 mg/g). (CONCLUSION) It is proposed that the best extration temperature and reagent was 40℃ and ethanol95%,respectively.
